Whoever secures the last spot will join a group featuring world No. 1 Jannik Sinner, Alexander Zverev, and Ben Shelton.
On Friday, Musetti is set to face American Sebastian Korda in the semifinals.
The top seed of the tournament, world No. 5 Novak Djokovic, is also aiming to reach the semifinals as he prepares to play Nuno Borges later on Thursday.
The 38-year-old Djokovic has already qualified for Turin and will compete in a group with Carlos Alcaraz, Taylor Fritz, and Alex de Minaur.
